Basic Principles:

  • Electric Field:
    • Electrophoresis involves applying an electric field to a medium containing the molecules to be separated.  
    • Charged molecules migrate through this medium towards the electrode with the opposite charge.  
  • Molecular Properties:
    • The rate of migration depends on the molecule's charge, size, and shape.  
    • Smaller, highly charged molecules move faster than larger, less charged ones.  
  • Supporting Medium:
    • A supporting medium, often a gel (like agarose or polyacrylamide), provides a matrix through which the molecules travel.  
    • The pores within the gel act as a sieve, further separating molecules based on size.  

Types of Electrophoresis:

  • Gel Electrophoresis:
    • This is the most common type, using gels as the supporting medium.  
    • Agarose gel electrophoresis:
      • Primarily used for separating DNA and RNA.  
    • Polyacrylamide gel electrophoresis (PAGE):
      • Commonly used for separating proteins.  
      • SDS-PAGE (Sodium Dodecyl Sulfate PAGE):
        • A variation that denatures proteins and separates them based on molecular weight.  

 

  • Capillary Electrophoresis:
    • Performed in narrow capillary tubes, offering high resolution and sensitivity.  
  • Isoelectric Focusing (IEF):
    • Separates proteins based on their isoelectric point (pI), the pH at which they have no net charge.  

Applications:

  • DNA Analysis:
    • Used in DNA fingerprinting, genetic testing, and forensic science.  
  • Protein Analysis:
    • Used to study protein expression, purify proteins, and diagnose diseases.  
  • RNA Analysis:
    • Used to analyze gene expression and study RNA structure.  
  • Medical Diagnostics:
    • Used to identify abnormal proteins in blood or urine, aiding in disease diagnosis.  
  • Research:
    • Essential in various research areas, including molecular biology, genetics, and biochemistry.

    • Maintains the pH and provides ions for conductivity.  
  • Gel:
